Abstract
Bioassays were carried out with two iodinated insulin preparations containing 0.8 and 3.0 atoms of iodine per molecule of insulin. In these preparations 80% and 30% of the original biological activity was retained.
With the help of the experimentally found reactivities of the tyrosine residues of insulin a statistical calculation is made of the iodine distribution over the insulin molecules at a certain (average) incorporation. On account of these distribution data it is reasonably certain that the substitution of one atom of iodine has no effect upon the biological activity. It seems likely that insulin molecules which contain two atoms of iodine also possess biological activity.
