✦ Synopsis

a-Glucosidases separated on isoelectric focusing gels can be localized by a two-step staining procedure. First, bromonaphthyl-cu-D-glucoside is cleaved by the a-glucosidases in the gel; then, the released bromonaphthol is precipitated with Fast Blue B salt. The procedure is specific for a-glucosidases, gives sharp enzyme bands, and does not inactivate the a-glucosidases. Using this procedure, it has been demonstrated that the maltase and a-methylglucosidase induced by maltose are separate enzymes.
