β¦ Synopsis
A simple method for the preparation of rat liver mate oxidase is described. The enzyme was purified from rat liver homogenate by cell fractionation, detergent treatment, alkali treatment, and affinity chromatography on %aminoxanthinebound Sepharose 4B. This enzyme preparation had a specific activity of 9.1 Uimg of protein and was purified about IOOO-fold from the liver homogenate. After sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis followed by staining with Coomassie brilliant blue, this preparation yielded one protein band at a position corresponding to a molecular weight of 33,000.
