A Sensitive Assay for Xanthine Oxidase Using Commercially Available [14C]Xanthine

A scaled-down radiochemical assay for xanthine oxidase using commercially available ['Vlxanthine is described. The substrate is purified by polyvinylpyrollidone column chromatography to decrease blank values. Inhibitors, if present, are removed from blood serum by gel filtration. Products are eluted from Dowex SOW and quantitated by liquid scintillation counting. Linearity was observed from 0.01 to 36.4 &r/sample.

The use of xanthine oxidase in immunoanalysis has never been reported. We describe here a procedure in which the xanthine oxidase dependent luminescence of luminol is enhanced in the presence of FeEDTA complex, providing an highly sensitive assay (3 amol of enzyme) and a long-term signal. This spec
