β¦ Synopsis
A simple, rapid method for detection of heroin and its metabolites in blood and urine is described. The technique employs alkaline hydrolysis in an autoclave for 15 minutes followed by two-dimensional TLC using ethyl acetate:methanol:ammonia (85 : 10 :5) and ethanol :dioxane :benzene :ammonia (5 :40 5 0 :5) as developing solvent systems. Visualization of spots is by ferric chloride/ferricyanide spray. Incomplete hydrolysis of the alcoholic 6-acetyl morphine metabolite was found, aiding reliable identification. No dealkylation of codeine occurred during the hydrolysis step.
