✦ Synopsis

We have developed a rapid, sensitive, and specific assay for photoreactivation of pyrimidine dimers in 3H-labeled DNA. It is based on the nuclease resistance of dimercontaining sequences in DNA, and the adsorption of these sequences to DEAE-substituted paper (DE-81). The method maintains the advantages of the rapid dimer assay of B. M. Sutherland and M. J. Chamberlin (1973, Anal. Biochem., 53, 168-176), while avoiding its major drawback, the frequent preparation of high specific activity, 32P-labeled, bacteriophage DNA. In addition. the use of scintillation spectrometry in our assay should allow more widespread use of this method.

## Abstract A ^3^H^+^‐release method has been developed for the assay of β‐hydroxylation of the adrenolytic drug mitotane. β‐^3^H‐mitotane was synthesized by the reduction of 1‐(2‐chlorophenyl)‐1‐(4‐chlorophenyl)‐2,2,2‐trichloroethane by an aluminium‐Hg~2~Cl~2~ couple in the presence of ^3^H~2~O. We have developed a rapid, highly sensitive method for the separation of thymine-dimers from thymine on a cation-exchange resin, which is capable of detecting one dimer out of 5 X 10" to 10" thymine molecules (depending on the specific activity of the used material) by the means of high-pressure liq
