✦ Synopsis

A pullulan-degrading enzyme activity was detected in the culture medium after growth of the fungus Aureobasidium pullulans on corn syrup as a carbon source. The product of pullulan catabolism by this activity was determined to be glucose. The highest specific activity of this enzyme was found after 7 days of fungal growth. Although a-amylase activity was not detectable, the presence of a glucoamylase activity was indicated. It appeared that the enzyme responsible for pullulan degradation was likely glucoamylase B.

A method to select lysine-excreting mutants from the fungus Aureobasidium pullulans was devised. This method involved the use of bacterial auxotroph indicator solid medium where mutant colonies were detected by their ability to support the satellite growth of an Escherichia coli lysine auxotrophic s
