✦ Synopsis


A novel positive/negative selection cassette, pu⌬tk, was generated. pu⌬tk is a bifunctional fusion protein between puromycin N-acetyltransferase (Puro) and a truncated version of herpes simplex virus type 1 thymidine kinase (⌬Tk). Murine embryonic stem (ES) cells transfected with pu⌬tk become resistant to puromycin and sensitive to 1-(-2-deoxy-2-fluoro-1-␤-D-arabino-furanosyl)-5-iodouracil (FIAU). Unlike other HSV1

tk transgenes, pu⌬tk is readily transmitted through the male germ line. Thus pu⌬tk is a convenient positive/ negative selectable marker that can be widely used in many ES cell applications. genesis 28:31-35, 2000.
