Homologous whole histone from calf thymus was adsorbed on Sepharose 4B columns with covalently coupled histone fractions H2a, H2b, H3 or H4 in 0.01 M phosphate buffer, pH 6.7 - 1 M NaCl. The adsorbed histones were eluted from the columns with 5 M urea in the same buffer. Electrophoretic analysis has

The synthesis of difluorescein disulfide, a new fluorescent thiol-specific reagent, is described. The absorption and fluorescence spectra and pH dependency of this compound and of several other fluorescein derivatives are presented. Histone H3 labeled with difluorescein disulfide displays a high flu
