✦ Synopsis

Clostridium thermocellum culture media containing crude cellulosic substrates, prepared by the steam-explosion of wood or straw, were found to interfere strongly with the Lowry method of protein determination.

Investigations into the mode of interferences demonstrated that either the intercept (blank value) and/or the slope (intensity of color development) were affected.

The efficiencies of precipitation procedures using the trichloroacetic acid method and the deoxycholate-trichloroacetic acid method were compared.

A modification of the method of hydroxyproline determination in proteins was devised. The modification consists of the hydrolysis of proteins in 72% perchloric acid of 100Β°C for 2 hr instead of 20 hr, autoclaving in 6 N HCl or 2 N Ba(OH)\*. Determination of hydroxyproline by the modified method does

Low levels of insoluble aggregates in protein formulations can sometimes only be detected by visual inspection. To overcome the subjectivity and other limitations associated with visual inspection, a microscopic technique based on filtration/staining was developed. This method is a simple modificati
