Wood et al. (1) investigated a tryptophanase preparation obtained from Escherichiu coli, and found that it converted tryptophan to indole, pyruvate, and ammonia, and that pyridoxal phosphate was needed as a coenzyme for the reaction. Utilizing this reaction, Wada et nl. (2) determined the amount of pyridoxal phosphate present in rabbit tissue and human serum by measuring the amount of indole produced, and McCormick et al. ( 3) simplified the method. The method presented here is similar to the macro method proposed by McCormick
(3) but has been modified and reduced tenfold from the original. This micro method has been applied to the determination of pyridoxal phosphate in both rat and human leukocytes and in rat liver.
