✦ Synopsis

INTRODUCTION. TEM studies of isolated cell cultures are performed after processing of the samples through a number of stages.

In oun laboratory, the fixed cells are initially packed in agar, collagen and other media rendering ensuing processing of the materials easier for embedding in epoxy resin. Using this method, a greater number of cells is initially required because of attrition in the intermediate stages.

Immobil ization of cells is applied in experiments with hepatocytes, Langerhans islets (1,3), hybridomas ( 2 ) , and other cell types.

The usage of such preparations contributes to elucidation of problems in growth regulation and cell differentiation.

Cells grown on type I hydrated collagen gels require special techniques for sample preparation and processing in order to optimize the removal of all reagents from the collagen matrix and prevent artifactual shrinkage. This method includes cutting out a small block of the collagen gel, postfixation,

The PVD preparation of thin, oriented films for electron microscopy of materials with high adatom mobilities on the substrates requires cooling of the substrate holder. The construction of such a holder for cooling the substrate to -50 and heating it to 500 degrees C is described.
