Polyvinylpyrrolidon und Methylzellulose reagieren mit Lokalanasthetika zu Komplexverbindungen. Die Komplexbildung mit Polyvinylpyrrolidon ist ausgepragter und wird vonviegend durch hydrophobe Wechselwirkungen und Wasserstoffbriicken verursacht. Das Ausmai3 der Wechselwirkungen ist bei Methylzellulose gering und wird durch Anwesenheit von Elektrolyten noch vermindert.
Die Bindung von Lokalanasthetika an Polyvinylpyrrolidon tr@t zur Verzogerung ihrer Freisetzung bei; der Depoteffekt ist jedoch vorwiegend auf die erhohte Viskositat der Zubereitungen zuriickzufuhren.
Interactions of Local Anesthetics with Macromolecular Additives and their Influence on Drug
Liberation Poly(vinylpyrro1idone) and methylcellulose react with local anesthetics to form complex compounds. Complex formation with poly(vinylpyrro1idone) is mainly due t o hydrophobic interactions and the formation of hydrogen bonds. Interactions with methylcellulose are weak. They decrease when electrolytes are added. Delayed release of local anesthetics is partly due t o their binding t o poly-(vinylpyrrolidone), but is mainly caused by the increase in viscosity.
