The constitutive heterochromatin regions of Callus domesticus were examined in mitotic cells of 16-to-18 hour embryos and meiotic cells of cockerels. Cbanding regions were discovered in almost all chromosomes including the macrochromosomes. These results differed from those of earlier reports. Most of the C-bands were located at centromeric and telomeric regions. Variability of C-banding in mitotic cells existed among embryos, among cells from the same embryos and between homologues. This variation, however, involved the frequency of occurrence of C-bands in a given location and was not the result of differences in the position of C-bands on a specific chromosome. The variable results were thought to be the consequence of differing amounts of DNA denaturation and extraction within a given C-band region. It was suggested that present-day C-band techniques may need refining to enable consistent detection of heterogeneous or small regions of constitutive heterochromatin. From the results obtained, an idiogram indicating the C-banding regions in Gallus domesticus was prepared. In addition, two C-band polymorphisms located in the Z-chromosome were reported.
