β¦ Synopsis
The use of 1 -fluoro-2,4-dinitrobenzene (FDNB) as a probe for the presence of hydrodisulfide groups in proteins is described. The technique involves derivatimtion of the protein with FDNB and subsequent treatment with dithiothreitol (DTT). The 2,4dinitrothiophenol
(DNTP) released from the PDNB-derivatized protein on treatment with DTT is determined spectrophotometrically. Using this technique, approximately 95% of the cyanolyzable sulfur of sulfurbound rhodanese was found to be released as DNTP. PDNBderivatized xanthine oxidase was found to release DNTP on treatment with DTT. However, no significant release of DNTP was detected on the treatment of either bis(2-amino-2-carboxyethyl)trisuffide (cystine trisuhide) or potassium thiomolybdate with DTT following incubation with FDNB.
