β¦ Synopsis
This paper describes a novel method of assaying urease (urea amidohydrolase, EC 3.5.1.5) which affords certain important advantages over other methods. This method utilizes titration with alkali and will therefore be called the "alkalimetric method." Also presented is a definition of the unit of activity in accord with hhe recommendations recently made by the International Union of Biochemistry (IUB) (1, 2). The companion paper that follows (3) reports some observations on the assay method of Sumner (4), which has been widely used in the past'. A factor for interconverting the results obtained by these two assays will be given. The present paper also reports values of the specific act'ivity found recently for highly purified urease preparations and summarizes pertinent earlier result's. MATERIALS

## Poly(adenosine diphosphate ribulose) [poly(ADP-Rib)] glycohydrolase activity was determined by measuring the amount of ADP-Rib hydrolyzed from polymers of ADP-Rib as substrate. In principle, the method consists of incubating oligomers or polymers of [W]ADP-Rib with testis glycohydrolase. The re
It has been shown that sulthydryl enzymes can be activated in a two-phase system by mercaptans which are coupled to Sepharose beads. Such an activator permits the use of thiol esters as substrates for enzymes requiring mercaptan activators, since the activator mercaptan and product mercaptan are eas
