✦ Synopsis
The quantity of extraeellular Na was descreased by 20°/0 in conscious dogs by peritoneal dialysis and mannitol infusion.
Balance studies and space measurements revealed a diminished intracellular sodium concentration which was the result of both sodium loss from cells and entry of osmotic free water.
Corresponding to these changes in sodium and water distribution a decrease in sodium excretion, measured as percent of filtered load, was noted.
Under these experimental conditions, changes in GFR to =~ 20°/0 and hypotonic ECF expansion did not alter the reduced rate of sodium excretion. No increase in sodium excretion was observed in osmotic diuresis or when spironolaetone was administered in the presence of lowered intracellular Na concentration. Finally, when intraeellular Na concentration was increased by means of changing from a low to a high salt diet, an increased sodium excretion was noted.
These observations suggest that the intracellu]ar sodium concentration is one factor influencing renal regulation of sodium homeostasis.
