Conventional chromatographic
procedures are inadequate for the separation of catecholamines for at least two reasons: (1) poor resolution and (2) decomposition of catecholamines into cyclic derivatives. Reaction of catecholamines with dansyl Cl (1-dimethylaminonaphthalene-5sulfonyl chloride) prior to chromatography probably prevents decomposition by dansylation of the phenolic hydroxyls and amino group. Dansylation of catecholamines also enhances chromatographic resolution and detectability.
Dansyl Cl was first introduced as a reagent for the N-terminal analysis of proteins and peptides by Gray and Hartley (1, 2)) who found that most dansyl amino acids could be separated using high-voltage paper electrophoresis at pH 4.4. Others (3-5) reported the separation of dansyl amino acids by thin-layer chromatography on silica gel and aluminum oxide using organic solvent systems. The separation of the dansyl derivatives of catecholamines by two-dimensional thin-layer chromatography was reported by Seiler and Wiechmann (6). We have developed a sensitive system for the separation of dansylated catecholamines by unidimensional ascending paper chromatography.
