✦ Synopsis
A fibre-ophc blosensor based on the fluonmetnc detectIon of NAD(P)H consumed or generated durmg enzymatuz reactions is described The design combmes a rnmlaturlzed fluonmeter Hnth a low-volume membrane reactor and utlhzes flow-mjectlon analysis prmclples Two types of membrane reactor were developed and tested In the first version, enzymes and a macromolecular mcotmanude adenme dmucleotide coenzyme derlvatlve of polyethyleneglycol, PEG-N6-(2-ammoethyl&NAJXP)(H), are retamed m the direct vxxmty of the sensor tip In the second version, a sohd-supported hquld membrane was used to retam native coenzyme, whde substrates and products were transported via selective tamers through an orgamc membrane barrier The automated determmatlon of phenylpyruvate, pyruvate, lactate, formate, ethanol and manmtol concentrations m model mecha was possible m the range of 50-1500 pm01 1-l The usefulness of the lxoscnsor concept reported for real process momtonng m tuotechnology or for medxal purposes was demonstrated on different examples mcludmg the deterrmnatlon of ethanol for Saccharomyces cerevrslae cultlvatlon
