Methods
Materials. F-12 Coon' s modification culture medium, all medium supplements, and fluorescent phalloidin were obtained from Sigma Chemical Company (St. Louis, MO). Fetal bovine serum was from Multiser (Cytosystems, Castle Hill, Australia). Geneticin (G418 sulphate) was purchased from GIBCO LTD (Paisley, Scotland). Rabbit polyclonal and mouse monoclonal antibodies directed against several apical (dipeptidylpeptidase IV [DPPIV], HA4, 5Π nucleotidase [5Π-NT], polymeric IgA receptor [pIgA-R]) and basolateral (CE9, HA321) plasma membrane markers 6 were generous gifts from A. Hubbard (Johns Hopkins University, Baltimore, MD). Rabbit polyclonal antibodies directed against the apical alkaline phosphodiesterase B10 and against ezrin were kindly provided by M.
