โฆ Synopsis
Degumming of crude cottonseed oil yielded 10/0 of phospholipids. Repeated acetone precipitations still left 18 o/o of triglycerides and 3.6 O/O of gossypol. The UV-spectrum of the total phospholipids showed a h,,, at 391 rnK w h i h was characteristic of the bound form of gossypol, possibly with the free amino group of the phosphatidyl ethanolamine present. The phospholipids were separated into a number of fractions by silicic acid column chromatography. Gossypol occurred in free form in the eluted fractions indicating cleavage during column chromatography. Analysis of the fractions by thinlayer chromatography as such and after different types of hydrolysis revealed the composition of the total phospholipids to be: phosphatidyl ethanolamine 22
O / o , inositol phospholipids 37 O / o , phosphatidyl choline 33 O / o and unidentified 8 O/o. Trotz mehrmaliger Fallung mit Aceton enthielt das gesamte Phospholipoid no& 18 o/o Triglyceride und 3.60/0 Gossypol. Das UV-Spektrum der gesamten Phospholipoidc zeigte ein Maximum bei 391 rnp, das fur gebundenes Gossypol charakteristisch ist. Diese Bindung entsteht wahrsheinlih mit der freien Aminogruppe des vorhandenen Phosphatidylathanolamins. Die Phospholipoide wurden uber Kieselgel saulen-chromatographisch in mehrere Fraktionen getrennt. Freies Gossypol war in den eluierten Fraktionen nahweisbar. Dies deutet auf eine wahrend der Saulen-Chromatographie eingetretene Spaltung hin. Die DC-Analyse der einzelnen Phospholipoid-Fraktionen und deren Hydrolyseprodukte ergab, daD das gesamte Phospholipoid aus 22 O/O Phosphatidylathanolamin, 37 O/O Phosphatidylinosit, 33 O/O Phosphatidylholin und 8 O / o n i h t identifizierter Stoffe besteht.
