Identifying the cellular rcsponsc< tc> photodynamic therapy (PDT) i h important if the mechanisms of cell death arc to be fully understood. PDT with a methylene bl ue analog DO15 yi el ded mitochondrial photodamage whilst membrane and lysosomal integrity were maintained. Apoptosih wax detected using the DNA stain HO342 by the appearance of 50 kb fragments and by DNA l adder formation. The release of mono-and oligonucleosomes was further quantilied using an ELISA protocol. Large DNA fragments were observed immediately l'ollowing illumination, and nucleosomes were detected at l-2 h post-treatment. increasing the dose 4-fold accelerated the apoptotic responvz to PDT. This i\ the first report of a thiuzine photoscnsitiser inducing apoptobis and is consistent with recent proposals suggesting that ~relcase of mitochondrial componcnt~ may play an irnporrant role i n the rnechanixin ot'cell death. 0 199X El\evier Science S.A.
