A comparative study was made of three scanning electron microscope (SEM) preparative techniques applied to protoplasts of the fern Pteridium aquilinum. All methods required removal of osmoticum and hence prior fixation, but single fixation, in glutaraldehyde or osmium tetroxide, or doublefixation, u
