The autologous mixed lymphocyte r e a c t i o n (AMLR) r e p r e s e n t s t h e p r o l i f e r a t i o n o f T c e l l s i n response t o s i g n a l s from autologous non-T c e l Is. F r a c t i o n a t i o n o f t h e non-T p o p u l a t i o n i n t o B e n r i c h e d and macrophage e n r i c h e d c e l l s demonstrated t h a t b o t h c o u l d serve as e f f e c t i v e s t i m u l a t o r c e l l s i n t h e AMLR. C y t o l y t i c t r e a t m e n t o f b o t h p o p u l a t i o n s w i t h a macrophage s p e c if i c , monoclonal a n t i b o d y abrogated s t i m u l a t i o n o f t h e macrophage b u t n o t t h e B c e l l p o p u l a t i o n . U t i l i z i n g a s e r i e s o f n e g a t i v e s e l e c t i o n proceduresc y t o l y s i s w i t h T c e l l s p e c i f i c monoclonal a n t i b o d y , m e t a b o l i c s u i c i d e with 5-bromo-2-deoxyuridine (brdU) and l i g h ti t c o u l d be demonstrated t h a t T c e l l s responding t o autologous macrophage were d i s t i n c t from t h o s e respons i v e t o autologous B c e l l s .

S t u d i e s o f t h e AMLR react i v i t y t o B c e l l s and macrophage i n a small number of p a t i e n t s w i t h a c t i v e systemic l u p u s erythematosus (SLE) demonstrated t h a t a l t h o u g h r e a c t i v i t y t o b o t h populat i o n s was diminished, t h e response t o autologous B c e l l s was reduced more t h a n t h e response t o a u t o l o g o u s macrophage.
