The synthesis of a fluorescein-derived fluorogenic substrate, D,L-myo-inositol 1- ([6'-hexyloxy-3-oxo]spiro[isobenzofuran-l(3H),9'-[9H]xanthen]-3'-yl hydrogen phosphate) sodium salt 9, for assay of activity of phosphatidylinositide-specific phospholipase C (PI-PLC), is described. This substrate is shown to be a sensitive substrate for bacterial PI-PLC in a continuous, non-radioactive assay.
