β¦ Synopsis
We report for the first time that subcellular morphologic information such as the size distribution of mitochondria can be noninvasively measured using polarized light spectroscopic techniques. An in vitro two-layer model was used to demonstrate the techniques, where the top layer consisted of mammalian cells (cervical or breast cancer cells) and the bottom layer was composed of a solid tissue-like phantom of Intralipid and India ink. The methods were first confirmed with 2.065 lm latex spheres as the top layer. A least squared optimization scheme coupled with Mie scattering theory was used to iteratively fit the measured intensity spectra. The results show that the size distributions of mitochondria obtained with our methods are in excellent agreement with that measured by fluorescence microscope.
