✦ Synopsis

A method for attaching water insoluble peptides to amine resins for sequence determination is presented. Derivitization of peptides with 4+ulfophenylisothiocyanate (s-PITC) greatly increased coupling efficiency. Conditions for removal of N-terminal s-PITC were investigated to maximize subsequent degradation of the resin bound peptides. Degradation of a resin bound, 24 residue peptide with thioacetylthioglycolic acid is described.
