Two-photon multicolor fluorescence in situ hybridisation (FISH) is presented as an advanced single molecule detection technique. Based on a two-photon excitation process it enables pinhole-free 3D laser scanning microscopy. Nonresonant two-photon absorption in a subfemtoliter excitation volume was r
