✦ Synopsis

The fluorimetric measurement of DNA at levels of 0. I-1.0 fig in cytoplasmic fractions of rat liver is described. The DNA was quantitatively precipitated from the cytoplasm derived from as little as 10 mg of liver with cetyl trimethyl ammonium bromide (CTAB) and the fluorescence produced after reaction with diaminobenzoic acid was measured. The precipitation was shown to give complete recovery of added calf thymus DNA or OH-labeled liver chromatin and the fluorescence produced was directly proportional to the amount of cytoplasm added. CTAB precipitation of DNA was also shown to be a useful substitute for phenol extraction prior to purification of DNA on hydroxylapatite columns.

## Abstract The purpose of this study was to investigate the problem of mitochondrial biogenesis in rat liver. The approach consisted of isolating mitochondria from control, 6 day starved and 6 day starved‐5 day refed rats and comparing their DNA, RNA and protein content. This was performed by isol

A method for the rapid, quantitative isolation of rat liver mitochondria has been developed. The method consists of placing portions of whole liver homogenate on linear Ficoll gradients followed by reorienting rate-zonal centrifugation in a vertical rotor. Fractions collected from the gradient were
