The fluorimetric measurement of DNA at levels of 0. I-1.0 fig in cytoplasmic fractions of rat liver is described. The DNA was quantitatively precipitated from the cytoplasm derived from as little as 10 mg of liver with cetyl trimethyl ammonium bromide (CTAB) and the fluorescence produced after reaction with diaminobenzoic acid was measured. The precipitation was shown to give complete recovery of added calf thymus DNA or OH-labeled liver chromatin and the fluorescence produced was directly proportional to the amount of cytoplasm added. CTAB precipitation of DNA was also shown to be a useful substitute for phenol extraction prior to purification of DNA on hydroxylapatite columns.
