โฆ Synopsis
Fig. 1. SDS polyacrylamide gel electrophoresis of homogenate supernatants (a) and incubation medium (b), after incubation of various tissues taken from the tarantula, Eurypelma californicum, with amino acids and 3H-leucine. Top: Gel stained with Coomassie Blue. Bottom: Fluorograph of the same gel treated with PPO [11], after 14 days exposure. ! Malpighian tubules, 2 pulmonary veins, 3 pericardium, 4heart wall, 5 'nascent' hemocytes, obtained by aspiration from inner heart surface. Lanes between tissue samples contain hemocyanin as reference. Note labelling of sample 5a only in the fluorograph [8, 9]. Although such cells have been examined by transmission electron microscopy (TEM) [10], their possible in- volvement in the hemocyanin biosynthesis has not been considered yet.
Failure to detect hemocyanin by TEM in spiders under normal conditions, may be due to its slow turnover when these cells are not activated. A specific stimulus, like blood loss, might force the cells to synthesiz e hemocyanin which then should also be revealed in their ultrastructure.
