β¦ Synopsis
ture antibody, B140 (5 l), was added to the resulting supernatant fraction (1 ml) and the Aβ€ content was analyzed using the method described above. The ratio of Aβ€ 42/Aβ€ 40 in the RIPA-soluble fraction was 0.073, which was similar to the ratio calculated directly (Aβ€ 40, 15.48 g/ml; Aβ€ 42, 1.08 g/ml) using anti-Aβ€ 40 and anti-Aβ€ 42 (Quality Controlled Biochemicals, Inc.) according to the method (8).
IP-MS offers a very convenient way to determine the ratio of Aβ€ 42/Aβ€ 40 and other Aβ€ peptide species in biological samples and should be a powerful and useful method for research into the pathogenesis of AD.
