Methionine-containing peptides are selectively isolated from complex digests by taking advantage of successive changes in hydrophobicity and chromatographic mobility induced by S methylation and demethylation of methionyl residues. The peptides are first separated by highperformance liquid chromatography in their S-['4C]methyl forms and then demethylated to facilitate their separation from contaminants during subsequent rechromatography.
