In Part I of this series (1)) Jonsson, Pettersson, and Rilbe have described the use of a commercial standard spectrophotometer cuvet (flowthrough cell, type 167-&S, made by Hellma GmbH, Miillheim, Baden, Germany) for density gradient isoelectric focusing of proteins on a microscale. By scanning the cuvet at intervals, the isoelectric positions of protein components can be registered, and the effect of prolonged electrolysis on the proteins can be studied. The isoelectric points of the separated proteins can be estimated by measuring the pH of isolated protein zones.

In this report, a method for fractionation of the cuvet contents into microfractions (60 ~1) is described, as well as the technique of measuring pH of the fractions. If desired, these fractions also allow estimation of possible biological activity.
