The conformation of DNA in alcohol solution has been studied recently using the technique of circular dichroism.14 It is accepted that DNA adopts its C-form in methanol in the presence of 5% SSC buffer.* However, ideas about the predominant conformation in a mixed solvent of methanol and ethanol (1:l + 5% buffer) are c ~n f l i c t i n g . ~, ~ Johnson and Girod3 concluded that DNA was completely denatured, whereas K a 9 proposed a retained doublestranded structure.
