P-815 mastocytoma cells increase the level of pyruvate kinase (PK) expression in response to c.hlorotorm-methanoI extracts of conditioned media, butyrate, and dibutyryl c-yclic AMP (but2cAMP) plus theophylline. The butyrate effect is indomethacin sensitive, suggesting a prostaglandin (PC) is the active signaling factor. Moreover, the chloroform-methanol extracts contain PGEz and PCF?,, and additions of the latter enhance PK artivity. PGE2 alone has little or no effect but acts synergistically with PCF,,. These data show that PCFL,, can regulate PK levels. On the other hand, other factors may also be active, since the end og en eou 5 and the b u t >CAM P p I u s t h eo p t i y l1 i n e e f tec t s a re i n d o m e t hacin insensitive. Most ot the factors that increase PK activity also inhibit cellular growth; however, regulation of PK expression tan be uncoupled trom growth in hi bi tion.
