β¦ Synopsis
The technique developed by Husek and modified by this laboratory for the derivatization of amino acids in aqueous solution using ethylchloroformate/trifluoroethanol/pyridine prior to gas chromatography positive-and negative-ion chemical ionization mass spectrometry was extended to the analysis of a broad range of dipeptides with diverse amino acid side chains. Twelve dipeptides and one tripeptide in aqueous solution were successfully analyzed with this technique. After isolation of a peptide mixture, the derivatization and analysis time is approximately 10 min. The detection limits of the dipeptide derivatives were generally in the picomole range. The method was also applied to the analysis of a commercial sugar substitute which contains a dipeptide and free amino acids.
