Acute barbiturate poisoning is now a common complaint.While 20 years ago the number of cases of poisoning from soporifics in Denmark amounted to about 100 per year it has now risen to 2000, most of which are caused by the intake of barbiturates (HEINTZELMAN and LINDILARDT 1949; Lous et al. 1949; WIINGAARD 1949).A clinical method for the determination of barbiturates in blood would, therefore, be of considerable value first and foremost in the differential diagnosis of patients suffering from unconsciousness. It would also give an indication of the extent of the poisoning, and valuable information would be gained during the course of the treatment. Further, it should be mentioned that a quantitative determination of barbiturates in the body fluids and tissues is essential for the elucidation of the yet unsolved pharmacological and pathophysiological problems concerning these poisonings.For several years methods which have been satisfactorily used in phorensic chemical investigations have been available, but these have not been applied to clinical work as they require large quantities of material and much time (SIMESEN 1939 ; HALSTRDM 1940 ; TURTITT 1948).
