Qzcantitative dctevmination of beef heart wiitochondria nztcleotides using a liqzcid ckromatograw
A procedure has been developed for the quantitative and qualitative estimation of nucleoside-5'-mono-, -di, and -triphosphates, nicotinamide and flavine nucleotides and nucleotides of .beef heart mitochondria. The nucleotides were separated by ionexchange chromatography on 0.6 x IO cm Dowex I X3 (16-20 p) columns, Cl-form, using a concentration gradient of HCl-NaCl. The procedure gives a high degree of resolution of nucleotides. The analysis was performed at 250, under a pressure of 8 atm, at a flow-rate of 120 ml/h and it was continued for 63 11. The total content of nucleotides of beef heart mitochondria was found to be 7.60 & 0.36 ,umoles per gram of protein, i.e. the AMP + ADP + ATP content was 4.76 & 0.30 pmoles, NAD+ + NADH content 1.47 & 0.16 ,umoles, the NADl?+ + NADPH content 0.64 &-0.09 ,umoles, and the FMN + FAD content 0.71 & 0.03 ,umoles per gram of protein.
