✦ Synopsis

Abstract

Mouse oocytes were found to sythesize proteins actively at the germinal vesicle, metaphase I, metaphase II, and pronuclear (6 hours post‐fertilization) stages. The qualitative pattern of protein components being sythesized in vitro, as demonstrated using polyacrylamide gel electrophoresis, changed throughout maturation and fertilization. Oocytes were arrested at metaphase I by β‰₯ 0.1 ΞΌg/ml cycloheximide or actinomycin D. The protein pattern in oocytes cultured in the presence of actinomycin D progresses to a metaphase II pattern in spite of the nuclear maturation arrest, indicating a dissociation between meiotic maturation and the changes in the pattern of proteins synthesized at different stages of maturation.

Mouse oocytes were previously observed to undergo structural changes involving the perivitelline space (PVS) within the oviduct following ovulation, as visualized by staining with fluorochrome-protein conjugates. Characteristic changes in the patterns of protein phosphorylation occur during meiotic maturation of mouse oocytes from the time subsequent to germinal vesicle breakdown, through metaphase 11, and following fertilization. These changes occur during both in vitro or in vivo maturation or fertilizatio

In animal oocytes, the centrosome exists as an acentriolar aggregate of centrosomal material that is regulated in a dynamic manner throughout the process of meiotic maturation. Recently, it has been demonstrated that in female meiotic systems spindle assembly is likely regulated by chromosomal and m
