A photosystem II enriched submembrane fraction was used in a photoelectrochemical cell kept in potentiostatic mode. We were able to generate strong photocurrents (55 PA cm-*) in optimal conditions, which included the presence of artificial electron acceptors acting as charge transfer mediators between the photosynthetic membrane and the working electrode. Electron acceptor concentration, pH and temperature affected photocurrent generation and oxygen evolution to the same extent. The electrochemical cell provided an adequate method for accurate electron transfer monitoring even when small quantities of photosystem II preparation were used.
