A gold-Con A probe w a s prepared u s i n g 10-15nm gold p a r t i c l e s (DeMey, 1984, EMSA B u l l e t i n , 1 4 : 5 4 ) . Sperm were l a b e l l e d w i t h t h e gold-Con A and s i l v e r i n t e n s i f i e d (Holgate, 1983, J Histochem Cytochem, 31:938), d i r e c t l y on t h e s l i d e s , which were shadowed w i t h v a r i o u s heavy metals and c o a t e d w i t h carbon. Some of t h e s l i d e s were coated w i t h a formvar or c o l l o d i o n f i l m b e f o r e immersion i n KOH d i g e s t i n g solut,ion.
