✦ Synopsis

The infusion of anti-idiotypic antibody in patients with lymphoma has proved a relatively innocuous procedure, but in general has yielded only partial, short-lived remissions of disease. A major problem is that xenogeneic antibody is simply not well-fitted t o destroying mammalian cells: complement and cellular effectors (K cells and phagocytes) are not efficiently recruited, and the target cells in any case present some excellent defense mechanisms. Antigenic modulation i s particularly prominent among these defenses, and we present evidence here for modulation of idiotype being much more efficient in vivo than in vitro. Two broad types of antibody derivative are under development t o improve the killing of neoplastic targets. One type relies on recruiting natural effectors, and is exemplified by univalent chimeric antibody. The other relies on delivering an exogenous effector such as a drug o r toxin, and i s exemplified by bispecific anti-ldlanti-saporin F(ab'r)l antibody. Both types of derivative have been able to suppress animal lymphoma t o the extent that tumor escape occurs largely through the emergence of idiotype-negative mutants.

## Abstract The enzyme specificity and tissue distribution of zenarestat, an aldose reductase inhibitor, was investigated in diabetic rats. Zenarestat inhibited both rat sciatic nerve aldose reductase (AR) activity and recombinant human AR with IC~50~ values of 7.5 and 44 nM, respectively. However,

## Abstract During the last decade, a central role for insulin‐like growth factor 1 (IGF‐1) in the pathophysiology of multiple myeloma (MM) has been well established. IGF‐I provided by the tumor–microenvironment interaction may directly and indirectly facilitate the migration, survival and expansio
