Determination of Total Nucleic Acids in Plant Tissue
Determination of nucleic acids in plant material is often more difficult than in animal tissues because of the high concentration of carbohydrates and ultraviolet-absorbing materials that interfere with procedures designed to measure the sugar moeity of the nucleic acid, or the direct measurement of ultraviolet absorption. These problems are especially acute in stem tissues, in which ratios between cell wall (85% carbohydrate) and total nucleic acid may be as high as 9: 1 (1).
Of many methods published (2, 3) the three most commonly used techniques are modifications of those originally described by Schmidt and Thannhauser (4)) Schneider ( 5)) and Ogur and Rosen ( 6)) and while no one method has achieved general recognition some are more favored for certain tissues than others.
