โฆ Synopsis
Consistent with our previous (A 2A KO) data in this dyskinesia model (Fredduzzi et al, 2002), the present study also showed that L-DOPAinduced dyskinetic movements were significantly attenuated in a conditional (Cre/loxP system) KO mice lacking postnatal forebrain A 2A receptors. Hence, both pharmacological and genetic data provide evidence that the A 2A receptor plays an important role in the development of sensitization to L-DOPA, and strengthen the rationale for targeting early PD in future clinical trials of A 2A antagonists in order to reduce the risk of developing dyskinesias.
