OBJECTIVE : To ohamotorixe ohoriouic relaxatiou against hypoxia. To mvestigato the e&t of aoute hypoxia rm placental vessels from patients-with severely abnormal uterine and umbilical Doppler study versus control group. MATERtAt AND METHODS:, 20 and 53 chorionic vessels from abnormal and normal ptaoeutas respectively were dissected. The vt%W were cquilibratcd in a Bicarbonate Kmbs ssltltio~~n~uously, bubbled with 95 % 02 The vessels wera contracted by an isotonic potassium KrkL solution,md were washed with normal sodium Krebs solution. After this initial ri$meu Thromboxane-Ttic U46619 was added to keep bath cmtcentratiou of IO' M. After submaxiial tension was achieved, vessels were exposed to hypoxia (95 % N2 and 5 % CO2 ) for one hour. The relaxation response to hypoxia was ovaluatsd a8 the percent of change in:,:tbe isometric force. genemmd during hypoxia, compared with those developed duriug the U-466 19

. 8 -on, Statistical analysis was petfbrmed. BESULTS: R&xation was reduced in a 73 % in abnormal group. Under hypoxic conditions there was a mhaxatior of 18.2 % 2 1.5 SEM in normal group. In the abttod group there was arelaxation of 4.9 %& 1.6 SEM ( pcO.65). CONCLUSIONS: A amdition of chronic hypoxia qay play a role impairing the capacity of pbtcental vasoulatme to reiav in response to acute hypoxia. Tbs mechanism invobwd in this phenomenon remains to be elucidated.
