Tubes with an O-ring joint have been dc~scribed (I, 2) for protein hydrolyses in place of sealed tubes. This technique eliminates the sealing of tubes and their troublesome opening. Also tubes wit.11 O-ring joints may be used repeatedly. Previous O-ring joint aplxuxtus have been modified in several respects and the prcscnt technique has been compared with the sealed-tube method with the hydrolysis of scrinc and threonine. Features of the apparatus include: ( 1) a Teflon stopcock sealed to an O-ring connector tube, (2) a new stopcock scaling clamp to press the Teflon plug in the stopcock, (3) use of a Teflon O-ring in the O-ring joint', and (4) a new O-ring joint sealing clamp.
