A technique which will separate superhelical, intact nonsuperhelical, and damaged mitochondrial DNA molecules by use of sedimentation gradients of formamide-sucrose is described. This technique can be used to detect damage to the mitochondrial DNA helix. @ 1987 Academic Press, Inc.
