Tetracycline forms chelate complexes with cupric, nickelous, and cob-& tous ioti that bind DNA when analyzed in a filter-retention a&y. Tetraz cycline complexes with other metal ions including zinc, ferrous, ferric, manganous, magnesium, and ca@ium ions do not pro_+ce f&is effect_ The tetracycline-copper complex binds the homopolymers pqlyriioadenylate, polyribouridylate, polyriiinosinate, and polyribocytidylate. The binding of polyribocytidylate is lea& effective. The preaddition of Cal& ions interferes with the ability of tetracycline to fo:m a DNA-binding complex with cupric ions. Calcium ions do not block DNA binding_ by an-already formed tetracycline-copper comp&x. Riboflavin interferes with the DNA-binding action _bf tetrac+clihe in the fWer-retention a~y.~ This suggests a rationale for its rep&ted antagonism of bacterial growth inhibition by tetr&ycline_ Riiofl& d&s not block the DNA-binding activity of an already formed te&acy@ne-copper c&npl&. None of the riboflavin-metal ion complexes tested are capable of binding DNA in *be filter-retention assay.
