✦ Synopsis
Reduced pyridine nucleotide and ammonia are required in equimolar amounts for the enzymic synthesis of glutamate from cr-ketoglutarate. Thus ammonia can be determined spectrophotometrically using NADH, a-ketoglutarate, and NADH-dependent glutamic acid dehydrogenase. With this reaction, the activity of ammonia-generating enzymes can be followed in a coupled optical enzyme assay. A procedure is presented for the determination of urease activity developed for bacterial cell-free extracts.
