Abstract
Platinum nitroimidazole complexes are potentially useful for sensitizing tumour cells towards radiation damage. We have studied reactions of cis‐[Pt(II)(MNZ)~2~Cl~2~], where MNZ is 1‐(2′‐hydroxyethyl)‐2‐methyl‐5‐nitroimidazole (metronidazole), with guanosine monophosphate (5′‐GMP) and the oligonucleotides GpC, GpG and d(GpCpGpC) using ^1^H NMR spectroscopy. The complex [Pt(MNZ)~2~(5′‐GMP)~2~]Cl~2~ · 7H~2~O was isolated and characterized. Novel syntheses of cis‐ and trans‐[Pt(^15^N‐DMZ)~2~Cl~2~], where DMZ is 1,2‐dimethyl‐5‐nitroimidazole (dimetridazole), and NMR spectra of ^15^N‐enriched intermediates in the ligand synthesis, and of these complexes are discussed. Reactions of the enriched cis complex with 5′‐GMP were investigated by NMR spectroscopy.
